What Semax is
Semax is a seven-residue synthetic peptide built from the ACTH(4-7) fragment with a C-terminal Pro-Gly-Pro extension. The literature describes it as lacking the hormonal activity of the parent ACTH molecule, which is the point of the modification.
Published research is largely preclinical neuroscience — rodent models examining cerebral ischemia, neurotrophin expression and neuroinflammation, with more recent work on copper binding and amyloid interactions. Human evidence exists but is limited and concentrated in a regionally specific clinical literature.
There is no FDA- or EMA-approved Semax product. Material supplied under this name is a research compound for laboratory use only.
The modified analogue problem
The most common identity issue with Semax is not adulteration but substitution with a close relative. N-acetyl Semax amidate is a distinct compound, separately indexed in chemical databases, sitting roughly 41 Da heavier than Semax as a result of its acetyl and amide modifications.
Both are legitimate compounds. The question a certificate has to answer is which one is in the vial — and a purity percentage cannot, because each could be individually pure. Only identity determination by mass or sequence resolves it.
A 41 Da separation is comfortably within the resolving power of ordinary mass spectrometry, so this is not a hard analytical problem. It is simply one that goes unanswered whenever a certificate reports purity alone.
The reason the modified analogue exists at all is instructive. Terminal modifications of this kind — acetylation at the N-terminus, amidation at the C-terminus — are a standard medicinal-chemistry approach to slowing enzymatic degradation, since the unmodified termini are where peptidases tend to begin. So the acetylated amidated form is not a corruption of Semax; it is a deliberate derivative with different properties. That is precisely why the distinction has to be made analytically rather than assumed from a product name: two legitimate compounds, closely related, sold under confusingly similar labels.
Reading the analysis
Confirm the reported mass is consistent with Semax near 814 Da rather than the acetylated amidated analogue. Semax contains a methionine residue, so it is one of the sulfur-containing peptides in common supply — a detail that can corroborate a formula on a certificate.
That methionine also means oxidation is a plausible degradation route, appearing as species 16 Da heavier. A certificate reporting related substances against a specification is describing this properly; one reporting a single number is not.
Beyond identity, apply the standard reading: purity by RP-HPLC with method and wavelength stated, net peptide content where reported, contamination controls present, lot matching the vial, and the document verifiable with the issuing laboratory.
One contextual note belongs on any honest page about this compound. A meaningful share of the human clinical literature on Semax originates from a single national research tradition and is not widely replicated elsewhere, which is a different evidential situation from a compound with multi-centre international trials behind it. That is an observation about the evidence base, not a judgement of the individual studies. It matters here because it explains why a compound can have decades of published work and still sit outside approved use anywhere — and why the analytical question of what is in the vial is the part a supplier can actually answer.

No. They are separately indexed compounds. N-acetyl Semax amidate carries acetyl and amide modifications that make it roughly 41 Da heavier than Semax. Both may be legitimate materials, but they are not interchangeable, and only identity testing on the certificate tells you which was supplied.
What mass should Semax show on a COA?
Approximately 814 Da for the seven-residue sequence. A result around 855 Da is more consistent with the N-acetyl amidated analogue, and species 16 Da above the expected mass are consistent with oxidation of the peptide's methionine residue.
Why is my Semax powder off-white rather than white?
Appearance is recorded on certificates as an observation, and slight colour variation can reflect residual solvent, moisture or oxidation rather than a different compound. It is a prompt to check the analytical data, not a determination in itself — colour cannot establish identity or purity.
Is Semax an approved medicine?
Not in the United States or European Union. There is no FDA- or EMA-approved Semax product. The published research base is predominantly preclinical, with limited human clinical literature, and material supplied for research is not a medicine.
